چکیده

Abstract Objectives Telomerase Cajal body protein 1 (TCAB1) is closely related to the occurrence, development and prognosis of tumors, may affect sensitivity tumor radiotherapy. There are no reports about effect TCAB1 gene expression on proliferation apoptosis HEPG2 cells. We plan investigate whether silencing using siRNA helpful for diagnosis treatment hepatocellular carcinoma. Methods Three sequences (siTCAB1-1, siTCAB1-2, siTCAB1-3) targeting one negative sequence (NC) were designed, synthesized then transfected into cells, separately. The expressions telomerase reverse transcriptase (TERT) in mRNA level detected by RT-qPCR Western blot assays. Moreover, cells measured MTT assay, flow cytometry, respectively. Results data both showed that siTCAB1 group was significantly lower than blank control NC groups (p < 0.05). However, TERT not different among those > result apoptotic rate increased compared with 0.01). Conclusions Silencing can inhibit promote providing a potential therapeutic method

Downregulation of Kinesin Spindle Protein Inhibits Proliferation, Induces Apoptosis and Increases Chemosensitivity in Hepatocellular Carcinoma Cells

Background: Kinesin spindle protein (KSP) plays a critical role in mitosis. Inhibition of KSP function leads to cell cycle arrest at mitosis and ultimately to cell death. The aim of this study was to suppress KSP expression by specific small-interfering RNA (siRNA) in Hep3B cells and evaluate its anti-tumor activity. Methods: Three siRNA targeting KSP (KSP-siRNA #1-3) and one mismatched-siRNA (...

Toosendanin inhibits hepatocellular carcinoma cells by inducing mitochondria-dependent apoptosis.

Toosendanin, a triterpenoid derivative isolated from Melia toosendan Sieb. et Zucc., possesses different pharmacological effects in humans and an important value in agriculture. As indicated by previous reports, the molecular mechanisms of toosendanin's anticancer effects remain poorly clarified. In this study we used both in vivo and in vitro models to investigate the anti-cancer effects of to...

TSG101 Silencing Suppresses Hepatocellular Carcinoma Cell Growth by Inducing Cell Cycle Arrest and Autophagic Cell Death

BACKGROUND The tumor susceptibility gene 101 (TSG101) was originally identified as a tumor-suppressor gene that mediates many molecular and biological processes, such as ubiquitination, endosomal trafficking, cell survival, and virus budding, but its role in hepatocellular carcinoma (HCC) is currently unknown. MATERIAL AND METHODS We assessed the expression of TSG101 in HCC and paracancerous ...

miR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting

Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...

FH535 suppresses the proliferation and motility of hepatocellular carcinoma cells.

The Wnt signaling pathway is activated in hepatocellular carcinoma (HCC). This study investigated the effects of FH535, an inhibitor of the Wnt signaling pathway, on the proliferation and motility of HCC cells. HLF cells and PLC/PRF/5 cells, HCC cells, were subjected to 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt (MTS) assay with the addi...
